| Brief outline of
the project : Mucosal epithelial cells constitute an
integral part of the host innate immune system and modulate the
development of the adaptive immune responses. Owing to the
unique disposition of the intestinal mucosal epithelial cells (IECs),
the most differentiated cells line the gut lumen and come in
contact with the luminal pathogens first and foremost. It is
widely believed that differentiation of the IECs plays a key
role in intestinal homeostasis and fighting off the pathogens to
invade human body. Expression of several receptors and effector
molecules that govern the gut immune response is modulated by
IEC differentiation. For example, antimicrobial peptides like
cathelicidin (hCAP18 / LL-37) and human defensin-1 are
significantly upregulated in sodium butyrate differentiated IECs.
We have recently reported that bacterial ADP-ribosylating toxins
like cholera toxin and LT markedly downregulate LL-37 and HBD-1
expression in the differentiated IECs (Cellular Microbiology
2008, 10(12), 2520-2537). However, little is known how
differentiation regulates gene expression in the IECs. This
project intends to thoroughly study the molecular mechanisms
behind the regulation of the genes that play important roles in
the host innate immune responses by the process of epithelial
differentiation. As host immune response is also critical for
chronic inflammatory diseases and cancers, this study will help
in better understanding of the development and progression of
inflammatory bowel disease and intestinal tumours.
